Collection:

The deciduous teeth are first early teeth in the mouth. The first teeth (first the down central cutters and second the upper ones) appear during the 7th and the 9th month after the birth, respectively. The last deciduous teeth (2nd molars) appear at the age of 2 ½ years. There 20 deciduous teeth in total; ten on the upper jaw bone and ten on the down jaw bone. From the 6th year of life these teeth are being replaced gradually by the stable teeth. The completion of the stable teeth is carried out during the 12th and the 13th year of life. Then the stable teeth are 28, until the appearance of the third molars at the age of 17 till 20. Then the total teeth number is 32, 16 on the upper jaw bone and 16 on the down one. With a simple and totally painless procedure (with local anesthesia) the dentist extract the deciduous teeth that are about to fall and put them into the collection KIT.

The dental pulp sample is being coded with a unique barcode for its traceability both during the processing and after the cryopreservation. Sample processing is being carried out immediately upon receive with validated procedures in a completely closed system (biogenea® set). In particular, the mesenchymal stem cells are being isolated from cord tissue through enzymatic digestion and centrifugation. The whole procedure is being carried out in a laminar flow class A/B (Annex I - February 2008 EU guideline for the cGMP standards).

A cell count is being performed both prior to and after the procedure. Prior to the procedure a full mesenchymal stem cell test is being performed. The mesenchymal stem cell analysis is carried out using the automated flow cytometry analyzer . After the procedure completion, a small amount of the final product is being used for the final numerical counting of the progenitor cells, as well as their viability prior to the cryopreservation using flow cytometry SOPs protocol. A small amount of the sample is being cryopreserved and thawed especially for a viability test of the mesenchymal stem cells after the cryopreservation procedure. Using this procedure we can guarantee that the viability and the plasticity of the mesenchymal stem cells are intact during the cryopreservation. The viability test is being performed using the automated luminometric system, while the plasticity test is being performed using the Mesencult system. A small amount of the stem cell culture is controlled for the presence of aerobic and anaerobic germs using the automated blood culture system, while in parallel the automated genetic analyzer is used for the detections of HBsAg, as well as for Hepatitis C, HIV Ι/ΙΙ , Toxoplasma , Syphilis and CMV.

Long term Cryopreservation & Product Release:

Only after the completion of all the quality tests isolated mesenchymal stem cells are stored in temperatures reaching the -196C ( in these conditions all the metabolic procedures are stopped ) and document/certificate of cryopreservation is issued.
